Cellcontactdependent, or cell-contact-dependent, describes biological processes and signaling that require direct physical contact between cells or between a cell and its immediate surroundings. This contrasts with soluble, diffusible signals such as hormones or cytokines that act over a distance. In many systems, juxtacrine signaling relies on membrane-bound ligands or receptors, necessitating contact for signal transmission.

Key mechanisms underlying cell-contact-dependent communication include receptor-ligand interactions across adjacent cell membranes, adhesion via cell adhesion molecules, and specialized structures such as gap junctions and immune synapses. Notable examples include Notch signaling, which requires neighboring cells to present ligands that activate Notch receptors on the adjacent cell; cadherin-mediated adhesion that coordinates tissue architecture and morphogenesis; and gap junctions that permit direct exchange of ions and small metabolites between connected cells. In the immune system, the interaction between T cells and antigen-presenting cells often depends on sustained cell-to-cell contact to achieve activation and specificity. Some bacteria also exhibit contact-dependent behaviors, such as contact-dependent growth inhibition, where physical contact delivers growth-inhibitory signals.

The term is used in development, immunology, cell biology, and microbiology to distinguish processes that rely on immediate cellular proximity from those driven by soluble factors. Study methods include co-culture systems, microfabricated devices that control cell contact, and imaging techniques to observe contact-dependent events in real time. Understanding cell-contact dependence is essential for insights into tissue patterning, immune recognition, and intercellular communication across kingdoms.
