In biology, a receptor-ligand interaction refers to the binding of a ligand to a receptor, typically triggering a cellular response. Most interactions are non-covalent and reversible, enabling dynamic regulation. Receptors are proteins located on the cell surface, in the cytosol, or in the nucleus, and ligands may be neurotransmitters, hormones, drugs, or other signaling molecules.

Binding outcomes depend on the ligand type: agonists activate receptors to elicit a response; antagonists block receptor activity; inverse agonists reduce constitutive activity. Allosteric modulators bind to sites distinct from the primary (orthosteric) site, altering receptor conformation and influencing the effect of orthosteric ligands.

Key properties include affinity and specificity. Affinity describes how tightly a ligand binds, often expressed as a dissociation constant (Kd); higher affinity corresponds to a lower Kd. Kinetic parameters (association and dissociation rates) determine how quickly binding forms and breaks apart. Some ligands show functional selectivity, triggering preferred signaling pathways.

Receptor-ligand interactions underlie many physiological processes and are central to pharmacology. Major receptor families include G protein-coupled receptors, ion channels, receptor tyrosine kinases, and intracellular receptors. Regulation occurs through desensitization, internalization, and downregulation. In drug development, understanding binding helps design compounds with desired efficacy, selectivity, and safety, using binding assays and structural analyses to study interactions.
