Baittosurface

Baittosurface is a term used to describe the approach of anchoring a molecular bait onto a solid surface to capture specific targets from a mixture. The bait may be an antibody, nucleic acid aptamer, receptor, enzyme substrate, or other binding partner chosen for its affinity for the target organism or molecule.

The surface provides a platform such as glass, silicon, gold, or polymer-coated substrates. Immobilization is achieved through chemical linkers and chemistries, including silanization for glass, thiol-gold self-assembly, NHS-ester coupling to amines, click chemistry, or biotin-streptavidin capture. Orientation control can be important; strategies include using bifunctional linkers or Fc-tagging.

Applications include biosensors (optical, electrochemical, SPR), microarrays, diagnostic assays, environmental monitoring, and research into molecular interactions. The method enables rapid capture and detection of targets, sometimes allowing multiplexing when multiple baits are presented on distinct surface regions.

Advantages include specificity, compatibility with small volumes, potential for label-free detection, and the possibility of surface regeneration. Limitations encompass non-specific adsorption, variable binding kinetics, fouling, loss of activity upon immobilization, and challenges in achieving uniform orientation.
