Self-assembly is a process by which a disordered system of components forms an organized structure without external direction, driven by local interactions and the ambient environment. It is a bottom-up approach contrasted with top-down fabrication, and is central to chemistry, physics, and biology.

The mechanisms rely on thermodynamics and kinetics. Components interact through non-covalent forces such as hydrogen bonding, ionic interactions, van der Waals forces, and hydrophobic effects; dynamic covalent chemistry can enable reversible bond formation. Structures emerge as the system minimizes free energy, though kinetic barriers can trap metastable states.

Types include molecular self-assembly (DNA, peptides), supramolecular assemblies, block copolymer assemblies, and colloidal assemblies (micelles, vesicles, colloidal crystals). Common examples are DNA base pairing and DNA origami, amphiphilic molecules forming micelles and lipid bilayers, viral capsids, protein fibrils, and inorganic nanoparticle superlattices.

Applications span nanomaterials, drug delivery, biosensing, molecular electronics, photonic materials, and tissue engineering scaffolds. Advances seek greater control over defect formation, yield, and scalability, as well as robust operation across environments.

The concept emerged in the 20th century through contributions across disciplines; notable progress includes the development of DNA nanotechnology in the 1990s. Today, self-assembly provides a foundational strategy for bottom-up design of complex materials.
