Threestranded is an adjective used to describe molecules or structures composed of three strands. In science, the term is most closely associated with triplex forms of nucleic acids, where a third strand interacts with a duplex to create a three-stranded complex.

Triplex DNA forms when a single-stranded third strand binds to a DNA duplex via Hoogsteen or reverse Hoogsteen hydrogen bonds, usually in polypurine–polypyrimidine sequences. The third strand can be purine-rich or pyrimidine-rich, and binding generally occurs in the major groove. Depending on composition, triplexes may be stabilized by protonation or chemical modifications.

Third strands are often generated synthetically as triplex-forming oligonucleotides (TFOs) or with peptide nucleic acids (PNAs), allowing targeted recognition of specific sequences. In some systems, cellular factors may assist triplex formation or stabilization.

Biologically, triplex structures have been explored as antigene agents that can inhibit transcription or alter replication and recombination. In vivo formation is influenced by chromatin context and cellular conditions; stability at physiological pH remains a challenge. Research seeks modified nucleic acids with improved affinity, specificity, and delivery, including LNAs and PNAs.

Beyond nucleic acids, threestranded architectures appear in materials science as three-stranded supramolecular assemblies and nanostructures. The broader concept highlights how multistrand architectures expand possibilities in gene regulation, nanotechnology, and materials design.
