Probe conjugates are hybrid molecules in which a chemical or biological probe is covalently linked to a second component to enable detection, imaging, or interrogation of biological systems. The probe provides a measurable signal or activity, while the second component provides targeting, recognition, or localization functions. Commonly, probe conjugates pair a fluorescent dye, radiolabel, chemiluminescent substrate, or affinity tag with a targeting scaffold such as an antibody, antibody fragment, aptamer, peptide, nucleic acid, small molecule, or nanoparticle.

Types of probe conjugates include fluorescent probe conjugates used for microscopy and flow cytometry, radiolabeled conjugates for positron emission tomography or single-photon emission imaging, and enzymatic or activity-based probes that generate a detectable readout upon interaction with a biological target. Conjugation typically relies on well-established chemistries such as amide coupling (carboxyl to amine) using NHS esters or EDC, click chemistry (azide–alkyne cycloadditions), maleimide–thiol linkages, or disulfide exchanges.

Key design considerations involve the nature of the probe and targeting moiety, the choice of linker in terms of length, rigidity, and hydrophilicity, and the stability of the conjugate in physiological conditions. Performance depends on factors such as signal-to-noise ratio, quenching or enhancement effects, and potential steric hindrance that can affect binding or activity.

Applications span basic research, diagnostics, and translational medicine, including live-cell imaging, tissue staining, and in vivo tracking. Challenges include stability, specificity, manufacturing consistency, and, for clinical use, regulatory compliance.
