Azidealkyne is a term used to describe chemical motifs that contain both an azide group (-N3) and an alkyne group (-C≡C-). These motifs are central to click chemistry and enable modular assembly of molecules through azide–alkyne cycloaddition. The azide and alkyne can reside on the same molecule (intramolecular cyclization) or on separate partners (intermolecular coupling).

The most studied reaction involving azidealkynes is the azide–alkyne cycloaddition (AAC). Under copper(I) catalysis (CuAAC), an azide reacts with an alkyne to form a 1,4-disubstituted 1,2,3-triazole with high regioselectivity and good yields under mild conditions. In the absence of catalysts, thermal cycloadditions give mixtures of 1,4- and 1,5-disubstituted triazoles. Strain-promoted azide–alkyne cycloaddition (SPAAC) uses strained cyclooctynes to achieve copper-free cycloaddition, enabling use in biological settings where copper would be problematic.

Applications of azidealkyne chemistry are broad. The motifs are used to link biomolecules, polymers, and materials. CuAAC is a robust method for bioconjugation, surface modification, and the construction of functional materials, while SPAAC is favored when copper toxicity or sensitivity is an issue. The reaction is valued for high efficiency, broad functional group tolerance, and compatibility with aqueous and mild conditions, making it a staple of modern chemical biology and materials science.
