megaprimer
Megaprimer refers to a large DNA fragment generated by PCR that contains a desired mutation or modification and sufficient flanking sequence to anneal to a target plasmid. In megaprimer PCR mutagenesis, a first PCR produces the megaprimer by using primers that introduce the mutation. The megaprimer is then purified and used in a second PCR with a plasmid template and a pair of short primers. The megaprimer primes synthesis of the mutated plasmid strand, incorporating the mutation into the circular DNA. The original template is typically removed by a post-PCR digestion step (for example with DpnI) to enrich for the mutated plasmid, which is then transformed and screened.
