Immunoprecipitating refers to the act of performing an immunoprecipitation, a laboratory technique used to isolate a specific antigen from a complex mixture by exploiting the binding specificity of an antibody. The goal is to enrich, detect, or study the target protein or protein complex from cells, tissues, or industrial samples.

The typical workflow begins with a sample containing the antigen, such as a cell lysate or tissue extract. An antibody specific to the target protein is added to form an antigen–antibody complex. This complex is then captured by a solid support, commonly beads coated with Protein A or Protein G or by magnetic beads, which bind to the antibody. The bead–antibody–antigen complex is separated from the rest of the mixture, usually by centrifugation or magnetic separation, and subjected to multiple washes to remove nonspecific proteins. The target protein can be eluted from the beads under reducing or denaturing conditions and analyzed by methods such as SDS-PAGE, Western blotting, mass spectrometry, or functional assays. When studying protein interactions, the technique is extended to co-immunoprecipitation, where binding partners are captured along with the target under gentle conditions to preserve protein complexes.

Variants include direct IP, where the antibody binds to the target in the lysate, and reverse IP, where an epitope-tagged protein is captured using an antibody against the tag. Native (non-denaturing) conditions aim to preserve interactions, while denaturing conditions focus on confirming identity. Controls, such as isotype antibodies, no-antibody controls, and pre-clearing steps, are essential to assess specificity and background. Immunoprecipitating is widely used for enrichment, verification of protein identity, post-translational modification analysis, and exploration of protein–protein interactions.
