In conventional terminology, the correct terms are hypoblast and primitive endoderm, and they describe a transient embryonic cell layer with specific developmental roles.

Definition and origin

The hypoblast, or primitive endoderm, arises from cells of the inner cell mass after implantation in mammalian embryos. This layer lines the part of the developing blastocyst cavity that becomes the extraembryonic space. The hypoblast contributes to the formation of the yolk sac endoderm and participates in signaling interactions that help pattern the overlying epiblast, which forms the embryo proper.

Development and function

As development proceeds, the hypoblast differentiates and migrates to establish the extraembryonic endoderm. It plays a supportive role rather than contributing directly to the embryo’s body, guiding lineage specification in the epiblast and helping to organize early tissues and signaling environments crucial for subsequent gastrulation.

Molecular markers

Markers associated with primitive endoderm/hypoblast include GATA4, GATA6, and SOX17, with additional contribution from PDGFRA and related endodermal signatures in various species. These markers help distinguish hypoblast-like cells in embryoid bodies and stem cell-derived models.

Relevance

In stem cell and developmental models, researchers sometimes use the term hypoblastprimitive to denote progenitor states that give rise to hypoblast-like lineages. However, the standard terminology remains hypoblast or primitive endoderm. Understanding this lineage is important for studies of early embryogenesis, yolk sac formation, and epiblast–hypoblast interactions.
