Homodimeric describes a macromolecular assembly composed of two identical subunits. In proteins, this means two identical polypeptide chains associate to form a functional dimer. The two subunits may be produced from the same gene (as a homodimer of identical polypeptides) or from different transcriptional outputs that yield identical sequences.

Subunits interact via non-covalent contacts such as hydrogen bonds, ionic interactions, van der Waals forces, and hydrophobic contacts; covalent disulfide bonds can also stabilize a homodimer in some cases.

Dimerization can be essential for function: it can create or expose active sites, enable cooperative or allosteric regulation, or provide structural stability. Homodimeric proteins may bind ligands, DNA, or other proteins more effectively than monomers. In some transcription factors, dimerization of identical DNA-binding domains enables sequence-specific binding and regulatory activity.

Relation to other oligomeric states: homodimers are contrasted with heterodimers, which comprise two different subunits. Some proteins can exist in multiple oligomeric forms, including monomeric, dimeric, and higher-order assemblies, depending on conditions.

Evolutionary and practical notes: Homodimerization can arise from gene duplication and subsequent maintenance of identical interfaces. In biotechnology, engineered homodimeric fusion proteins or self-complementary domains are used to study assembly, increase potency, or regulate activity.
