Dimerization

Dimerization is the process by which two molecular entities, called monomers, associate to form a single chemical species known as a dimer. Dimers can arise through covalent bonds or through noncovalent interactions such as hydrogen bonding, electrostatic forces, or van der Waals forces. The formation is often reversible and depends on concentration, temperature, solvent, and the presence of catalysts or oxidants.

Dimerization is commonly categorized as covalent or noncovalent, and as homodimer or heterodimer. In a homodimer, two identical monomers bind; in a heterodimer, two different monomers bind. Covalent dimers are linked by a chemical bond, for example disulfide bonds between cysteine residues in proteins or radical coupling products. Noncovalent dimers rely on reversible interactions and are widespread in biology, where many proteins function as dimers; receptor proteins and transcription factors often require dimerization to become active.

The concept is also important in chemistry and materials science. In radical chemistry, two radicals can combine to form a dimer, affecting reaction pathways and product distributions. In polymer science, dimerization can influence volatility, reactivity, and crystallinity of small molecules and can serve as a preparative step or side process in synthesis.

Dimerization is described quantitatively by a dimerization constant, K_dim, defined as [dimer]/[monomer]^2 for simple two-component equilibria. It is typically temperature-dependent and can be driven or suppressed by changes in concentration, solvent polarity, or redox conditions. Analytical techniques such as NMR, UV–vis spectroscopy, and mass spectrometry are used to detect and characterize dimers.
