In the mevalonate pathway, GGPP is primarily synthesized in the cytoplasm of eukaryotic cells and is derived from farnesyl pyrophosphate (FPP) through the enzymatic action of geranylgeranyl pyrophosphate synthase. This enzyme catalyzes the condensation of FPP with an additional isoprene unit, typically provided by isopentenyl pyrophosphate (IPP) or dimethylallyl pyrophosphate (DMAPP). GGPP is also generated via the MEP pathway in plastids, bacteria, and archaea, where it is formed from the condensation of two molecules of IPP.

One of the most significant roles of GGPP is as a precursor for the synthesis of geranylgeraniol, dolichols, and various terpenoids, including carotenoids and some phytosterols. Additionally, it serves as the substrate for the prenylation of proteins, a post-translational modification that often targets small GTPases such as Rab proteins, Rho proteins, and some G-protein-coupled receptors. This modification is critical for the proper localization and function of these proteins within cellular membranes.
