In vitro gastrulationbased systems use pluripotent stem cells to model or emulate aspects of gastrulation. Gastruloids, embryoid bodies, and related organoid formats can exhibit symmetry breaking, germ layer specification, and early patterning without forming a full organism. These systems provide insight into early development, axis formation, and tissue interactions, and serve as platforms for studying congenital defects, drug effects, and morphogenesis mechanisms.

Applications include basic developmental biology, disease modeling, regenerative medicine, and screening for teratogenic compounds. Methodologically, gastrulationbased studies rely on 3D culture, extracellular matrices, microfabrication to control geometry, and precise modulation of signaling with growth factors and inhibitors, often coupled with live imaging and lineage reporters.

Limitations and ethical considerations are ongoing. In vitro models do not fully reproduce the complexity of in vivo gastrulation, show species-specific differences, and raise questions about embryo-like constructs. Researchers emphasize careful interpretation, standardized criteria for defining germ layer formation, and compliance with applicable guidelines.
