Gap junction channels, often referred to as connexons in vertebrates, are specialized intercellular connections that directly link the cytoplasm of adjacent cells. These structures play a critical role in facilitating the transfer of ions, small molecules, and electrical signals between neighboring cells, thereby enabling coordinated cellular activity. Gap junctions are composed of proteins called connexins, which assemble into hexameric units that span the plasma membranes of two connected cells. The function of gap junctions is tightly regulated by various factors, including phosphorylation, pH, and calcium levels. Dysregulation of these channels has been implicated in numerous pathological conditions, including cardiac arrhythmias, neurodegenerative diseases, and certain types of cancer. For example, mutations in connexin genes can disrupt intercellular communication, leading to impaired tissue function. Additionally, altered gap junction expression has been observed in tumor cells, where it may promote tumor progression or metastasis.
