Autoregulatory describes a set of processes by which a system regulates itself to maintain stability or consistent output in the face of changing conditions. In physiology, autoregulation refers to an organ’s intrinsic ability to preserve stable function, such as constant blood flow, despite fluctuations in perfusion pressure. The term also appears in molecular biology to describe feedback loops in which a gene or protein regulates its own expression.

In physiology, autoregulation is most clearly observed in the brain, heart, and kidneys. Cerebral autoregulation keeps cerebral blood flow relatively constant across mean arterial pressures roughly between 60 and 160 mmHg. Renal autoregulation maintains a stable glomerular filtration rate over a range of arterial pressures, aided by afferent arteriolar myogenic responses and tubuloglomerular feedback. Myogenic mechanisms respond to stretch, while metabolic factors such as oxygen, carbon dioxide, and pH influence vessel tone. Endothelial signals, including nitric oxide and endothelin, modulate vascular tone. The pulmonary circulation operates differently and does not rely on the same autoregulatory mechanisms as systemic organs.

Clinical relevance includes the vulnerability of autoregulatory systems to disease. Hypertension can shift autoregulatory curves to higher pressures, reducing tolerance to low blood pressure. Acute brain injury or stroke can disrupt cerebral autoregulation, increasing risk of hypoperfusion or hyperemia. Management in anesthesia and critical care often aims to keep perfusion within the organ’s autoregulatory range.

In genetics and cellular biology, autoregulatory circuits are feedback loops where a gene product regulates its own production. These circuits can be negative, stabilizing expression, or positive, amplifying it. Autoregulatory regulation is common across organisms and contributes to development, homeostasis, and rapid response to changing conditions.
