Thermoreceptors-free is a term used to describe tissues or organisms in which functional thermoreceptors are absent or non-operational. Thermoreceptors are sensory neurons and receptor cells that detect temperature changes, contributing to the sense of hot and cold and to thermoregulatory reflexes. In vertebrates, peripheral thermoreception is mediated by sensory neurons in the skin and mucous membranes, often expressing transmembrane receptor channels such as TRPV1 (heat) and TRPM8 (cool). The concept implies a lack of effective thermal sensing at the peripheral level, with downstream effects on perception and reflexes.

Causes and mechanisms: Thermoreceptors-free states can arise from genetic manipulation, such as knockout or silencing of thermosensitive channels or of nociceptive neurons. Chemical ablation (for example, capsaicin or resiniferatoxin) or surgical denervation can remove peripheral thermal sensing. In practice, complete removal of all thermosensitive inputs is uncommon due to redundancy and compensatory plasticity, and central nervous system circuits may adapt to reduced peripheral input.

Consequences: The absence of functional thermoreceptors typically impairs thermal discrimination and thermoregulatory feedback from the skin. Subjects may fail to perceive harmful temperatures, increasing the risk of thermal injury. Central processing may adapt, leading to reliance on other sensory modalities or internal cues, and pain or aversive responses to extreme temperatures can be altered depending on the affected pathways.

Applications and research: Thermoreceptors-free models help delineate the contributions of peripheral receptors versus central processing in temperature sensation and pain. They inform studies of sensory pathway organization, anesthesia and analgesia strategies, and the development of prosthetic or restorative sensory feedback. Ethical considerations are important in the creation and use of such models in animals and human-derived systems.
