SDCBP

SDCBP stands for syndecan-binding protein and refers to a human gene that encodes the adaptor protein known as syntenin-1, also called MDA-9/Syntenin. The protein is a cytoplasmic scaffold characterized by two PDZ domains, which enable it to bind a wide range of partner proteins and organize signaling complexes.

Syntenin binds to the cytoplasmic tails of syndecans (Sdc1 through Sdc4) and associates with various transmembrane receptors, cytoskeletal components, and trafficking factors. Through its PDZ domains, it recruits partners to coordinate receptor internalization, trafficking, and signal transduction. It also interacts with ALIX and components of the ESCRT machinery, contributing to exosome biogenesis and the selective packaging of cargo into extracellular vesicles.

Biological roles of SDCBP include regulation of cell adhesion, migration, and invasion, as well as modulation of signaling pathways linked to growth factor receptors and integrins. By organizing cargo at the plasma membrane and endosomal compartments, syntenin influences how cells respond to their environment and communicate with other cells via exosomes and microvesicles. Its activity positions it as a central hub in coordinating membrane trafficking with intracellular signaling networks.

Clinical and research relevance: elevated expression of SDCBP has been associated with metastatic progression in several cancers, including melanoma and breast cancer, and experimental disruption of SDCBP function can alter metastatic potential in model systems. Beyond cancer, syntenin is studied for its roles in inflammation, neural development, and intercellular communication, reflecting its broad involvement in cellular signaling and cargo sorting.
