RNaseinactivating refers to methods and reagents used to prevent RNA degradation by ribonucleases (RNases) during laboratory workflows. RNases are widespread and highly stable enzymes found in many environments, making RNA work particularly fragile. Effective RNase inactivation is essential for RNA isolation, reverse transcription, and other processes that require intact RNA.

One major approach is the use of RNase inhibitors—proteins that bind tightly to RNases and block their activity. Commercial RNase inhibitors such as RNasin, RNaseOUT, and SUPERase-In are commonly added to reactions to protect RNA. These inhibitors are designed to inactivate a range of RNases, particularly RNase A-type enzymes, and are typically active under physiological conditions.

In addition to inhibitors, RNase inactivation can be achieved by chemical denaturation. Buffers containing chaotropic agents (for example guanidinium thiocyanate) or strong detergents can denature RNases during cell lysis and RNA extraction, simultaneously releasing and stabilizing the RNA. Heat treatment may inactivate some RNases but is not universally reliable, as many RNases are resistant to moderate heating. Chelating agents like EDTA can inhibit certain metal-dependent nucleases but do not effectively inactivate all RNases.

Maintaining an RNase-free environment is also critical. This includes using certified RNase-free reagents, sterile tubes, dedicated equipment, and protective practices to minimize contamination from skin and other sources. Storage and handling of RNase inhibitors should follow manufacturer recommendations, as improper handling can compromise effectiveness.
