Ribonucleases are pervasive in nature and can be introduced by skin, hair, dust, or inadequately cleaned instruments. They are highly stable proteins that can withstand many standard sterilization methods, making designated RNase-free practices essential. Contamination can occur at any stage, from reagent handling to sample processing.

To achieve RNase-free conditions, laboratories use certified RNase-free consumables (tubes, tips, plates), dedicated workspaces, and RNase-free reagents. Surfaces are cleaned with RNase-decontaminating solutions, water is treated to be RNase-free (for example, using commercially certified RNase-free water or DEPC-treated water that is properly inactivated), and gloves are worn and frequently changed. Equipment and reagents are sometimes sterilized or supplied as ready-to-use RNase-free preparations. It is common to maintain separate equipment and storage areas for RNA work to minimize cross-contamination.

Verification of RNase-free status can involve routine testing with RNase detection assays or monitoring RNA integrity after processing, for example by electrophoresis or microfluidic analysis (RNA integrity score). Despite precautions, RNase-free status applies to the tested batch and does not guarantee absolute sterility over long periods; ongoing vigilance is required.
