Proconvulsants
Proconvulsants are substances that lower the seizure threshold, making an individual more susceptible to seizures. They achieve this by disrupting the delicate balance between excitatory and inhibitory neurotransmission in the brain. Excitatory neurotransmitters, such as glutamate, increase neuronal firing, while inhibitory neurotransmitters, like GABA, dampen this activity. Proconvulsants can either enhance excitatory activity or impair inhibitory mechanisms, thereby tipping the scales towards excessive, synchronized neuronal firing characteristic of a seizure.
Various classes of drugs can act as proconvulsants. These include certain antibiotics, antidepressants, antipsychotics, stimulants, and
The proconvulsant potential of a substance is a critical consideration in pharmacology and toxicology. It influences
