Phosphonation

Phosphonation is a chemical process that introduces a phosphonate group into a molecule, forming phosphonate esters or phosphonic acids. The phosphonate motif features a phosphorus(V) center bonded to oxygen and carbon or other substituents, typically written as a P(O)(=O)(OR)2 or related structure. Phosphonates are valued for their chemical stability and their role as bioisosteres of phosphate groups in biology and medicine.

Phosphonates can be prepared by several principal routes. The classic Michaelis–Arbuzov reaction converts alkyl or aryl halides with trialkyl phosphites to dialkyl phosphonates, creating a direct carbon–phosphorus bond. The Pudovik reaction adds dialkyl phosphites to aldehydes or ketones to furnish α-hydroxy phosphonates. Other methods include reactions of alcohols with chlorophosphonates to form alkyl phosphonates or coupling of phosphonyl chlorides with nucleophiles to form aryl or alkyl phosphonates. The choice of method depends on the desired substitutions and the target phosphonate.

Applications of phosphonation span organic synthesis, medicinal chemistry, and materials science. Phosphonates serve as stable mimics of phosphates in drugs and enzymes, with bisphosphonates such as alendronate used in treating osteoporosis. They function as chelating agents, enzyme inhibitors, flame retardants, and ligands in catalysis. In materials chemistry, phosphonate groups enable surface grafting and the design of functional coatings.
