Nitrolinoleic

Nitrolinoleic acid, also known as N–(2‑nitro‑1-alkenyl) linoleic acid, is a nitrated polyunsaturated fatty acid that belongs to the broader class of nitro fatty acids. Its chemical formula is C18H30O4N and the compound contains a 9‑nitro group attached to the second carbon of the linoleic backbone, giving it characteristic electrophilic and anti‑oxidant properties.

The molecule can be produced from linoleic acid by nitration in the presence of nitric acid or peroxynitrite, and it is also formed in vivo during oxidative stress when reactive nitrogen species interact with unsaturated fatty acids. Natural sources of nitrolinoleic acid include certain foods such as tomato oil, soybean oil, and processed meats, where cooking and industrial processing can generate nitrated lipids. Laboratory preparation typically involves controlled electrophilic nitration in organic solvents followed by purification through chromatography.

Biological studies have demonstrated that nitrolinoleic acid exerts anti‑inflammatory, vasodilatory and anti‑thrombotic activities. It can modulate endothelial nitric oxide synthase activity, reduce pro‑inflammatory cytokine production, and protect against oxidative damage in endothelial cells. In animal models, it has been shown to alleviate experimental colitis, prevent atherosclerotic plaque formation, and reduce ischemia–reperfusion injury. Mechanistic investigations suggest that nitrolinoleic acid acts as a “redoxlator,” reacting with reactive oxygen species and signaling proteins via nitrosothiol chemistry.

Because of its multiple pharmacological effects, nitrolinoleic acid is being explored as a lead compound for developing new therapeutics targeting cardiovascular disease, inflammatory disorders, and metabolic syndrome. Current research focuses on improving its stability, bioavailability, and delivery to target tissues. No major adverse effects have been reported in pre‑clinical studies, but further investigation is needed to establish safe dosage ranges for human use.
