Metüültransferaasinimelistes, also known as methyltransferase inhibitors, are a class of compounds that interfere with the activity of methyltransferases, enzymes responsible for transferring methyl groups (CH₃) to various substrates such as DNA, RNA, and proteins. These enzymes play critical roles in epigenetic regulation, gene expression, and cellular metabolism, making methyltransferases important targets for therapeutic intervention.

Methyltransferases are categorized into different families based on their substrate specificity, including DNA methyltransferases (DNMTs), histone methyltransferases (HMTs), and RNA methyltransferases (RNMTs). Inhibitors of these enzymes are being explored for their potential in treating diseases where abnormal methylation patterns contribute to pathology, such as cancer, neurological disorders, and metabolic diseases.

DNA methyltransferase inhibitors, such as azacitidine and decitabine, are approved for the treatment of myelodysplastic syndromes and acute myeloid leukemia. These drugs induce hypomethylation of DNA, leading to reactivation of silenced tumor suppressor genes and restoration of normal cellular function. Histone methyltransferase inhibitors, like those targeting EZH2 (a component of the polycomb repressive complex), are under investigation for their role in epigenetic silencing in cancer.

The development of methyltransferase inhibitors faces challenges, including specificity, toxicity, and resistance. Off-target effects can lead to unintended epigenetic alterations, while resistance mechanisms, such as upregulation of alternative enzymes or compensatory pathways, may limit therapeutic efficacy. Research continues to refine inhibitor design, improve delivery methods, and explore combination therapies to enhance their clinical potential.

Beyond oncology, methyltransferase inhibitors are being studied for neurodegenerative diseases like Alzheimer’s and Parkinson’s, where abnormal methylation is linked to protein aggregation and neuronal dysfunction. Additionally, they may have applications in inflammatory and autoimmune disorders, where epigenetic dysregulation contributes to disease pathogenesis. Ongoing preclinical and clinical studies aim to expand the therapeutic scope of these compounds.
