LuminalpH

LuminalpH is a conceptual technology platform for measuring pH within luminal compartments of living organisms, such as the gastrointestinal tract or urinary tract, using luminescent indicators. The aim is to provide real-time, spatially resolved pH information with minimal invasiveness and physical disturbance.

Principle: The core components are pH-responsive luminescent probes whose emission properties change with pH. These probes can be incorporated into swallowable capsules, implantable devices, or applied via endoscopic tools. Readout methods may be optical, including spectral shifts, intensity changes, or lifetime variations, and typically require calibration to account for tissue scattering and autofluorescence. Data can be collected by external detectors or integrated imaging systems.

Applications: In research settings, LuminalpH supports mapping pH gradients along the GI tract, monitoring drug release profiles, and studying pathophysiology related to luminal pH. In clinical contexts, it could aid diagnostics and treatment planning for conditions such as reflux, ulcers, or inflammatory bowel disease, as well as in pharmacology studies of pH-dependent drug formulations.

Development and challenges: The field integrates chemistry, materials science, and medical imaging. Key challenges include ensuring probe stability in luminal environments, biocompatibility, accurate calibration, motion artifacts, and regulatory considerations. As of now, LuminalpH remains a topic of ongoing research with demonstrations primarily in preclinical settings.
