In its first season the league comprised twenty clubs, with each club playing the others twice for a total of thirty-eight matches per team. The competition is divided into a regular season, followed by a playoff series that crowns a champion. The playoff system has been periodically revised, currently featuring the top eight teams in a knockout format that culminates in a final held at a predetermined neutral venue.

The league has developed a reputation for high‑intensity play and has been a significant contributor to the international talent pipeline. A substantial number of players from LeaguePro12 clubs have been selected for their respective national teams for major tournaments such as the Rugby World Cup and the Six Nations Championship.
