Klason

Klason refers to a method for determining the lignin content in plant material. Developed by Swedish chemist Peter Klason, this acid hydrolysis technique is a standard procedure in pulp and paper research and other fields dealing with biomass analysis.

The Klason method involves treating a sample of lignocellulosic material with concentrated sulfuric acid. The acid hydrolyzes the cellulose and hemicellulose components of the plant material into soluble sugars. Lignin, being resistant to this treatment, remains as an insoluble residue. This residue is then filtered, washed, and dried, and its weight is measured. The percentage of this insoluble residue relative to the original sample weight is reported as the acid-insoluble lignin content.

While widely used, the Klason method has limitations. It can sometimes overestimate lignin content due to the formation of artifacts or incomplete hydrolysis of carbohydrates. Conversely, certain wood components may be lost during the process, leading to an underestimation. Despite these drawbacks, the Klason method remains a valuable and cost-effective tool for rapid lignin determination, especially in comparative studies. Variations of the method exist, such as the "milled wood lignin" or "native lignin" isolation techniques, which aim to obtain more representative lignin samples but are more complex and time-consuming.
