JAK2Exon12Mutation refers to specific genetic alterations occurring within exon 12 of the Janus kinase 2 (JAK2) gene. This gene plays a crucial role in cell signaling pathways, particularly those involved in the development and proliferation of blood cells. Mutations in JAK2, including those in exon 12, are commonly associated with myeloproliferative neoplasms (MPNs), a group of blood cancers characterized by the overproduction of one or more types of blood cells.

While the JAK2 V617F mutation in exon 14 is the most prevalent, JAK2 exon 12 mutations are found in a subset of patients with MPNs, particularly those with polycythemia vera (PV) who are negative for the V617F mutation. These exon 12 mutations typically result in a constitutively active JAK2 protein, leading to aberrant signaling and uncontrolled cell growth. Symptoms associated with JAK2 exon 12 mutations can overlap with other MPNs and may include an elevated red blood cell count, an increased risk of blood clots, and sometimes splenomegaly.
