Hexoosi6fosfaatteja, or hexose-6-phosphates, are a group of phosphorylated hexose sugars in which a phosphate group is attached to the sixth carbon of the sugar ring. The best-known member is glucose-6-phosphate; others include mannose-6-phosphate and galactose-6-phosphate. These molecules are important cellular intermediates that link carbohydrate metabolism with other metabolic pathways.

Formation and interconversion: In cells, hexoses are phosphorylated by kinases. Glucose is converted to glucose-6-phosphate by hexokinase or glucokinase. Mannose can be phosphorylated to mannose-6-phosphate by enzymes in the hexose kinase family. Glucose-6-phosphate can be isomerized to fructose-6-phosphate by phosphoglucose isomerase, feeding into glycolysis and related pathways. Glucose-6-phosphate also feeds the oxidative branch of the pentose phosphate pathway through glucose-6-phosphate dehydrogenase. Mannose-6-phosphate serves a distinct functional role as a targeting signal that directs certain enzymes to lysosomes via mannose-6-phosphate receptors.

Functions and significance: Glucose-6-phosphate acts as a central metabolic hub, funneling carbons into glycolysis, the pentose phosphate pathway, glycogen synthesis, or hepatic glucose production. Mannose-6-phosphate is notable for its role in trafficking lysosomal enzymes to lysosomes, which is essential for proper lysosomal function. The phosphate at the sixth carbon increases the molecule’s polarity, influencing its cellular localization and flux between compartments.

Clinical and research notes: Alterations in hexose-6-phosphate metabolism or trafficking can contribute to metabolic disorders or lysosomal storage conditions.
