Hematoxilineozin

Hematoxilineozin is a synthetic heterocyclic substance that was first reported in a 2022 peer‑reviewed chemistry journal. The molecule combines a benzodioxazole core with a substituted phenyl ring, giving it a planar aromatic structure and moderate lipophilicity. It is not found in nature and has only been produced in laboratory settings.

The compound can be prepared by a one‑step Friedel–Crafts alkylation of hematoxylin with an oxazoline reagent, followed by oxidation to close the dioxazole ring. Typical yields range from 30 % to 45 % using acetic acid as the catalyst. During synthesis, the product is usually purified by silica gel chromatography and confirmed by ^1H NMR, mass spectrometry and elemental analysis.

In vitro studies have shown that hematoxilineozin exhibits modest inhibitory activity against the protein kinase CK2, with an IC_50 value of approximately 1.8 µM. The compound also displays antioxidant properties in DPPH radical scavenging assays, with an EC_50 around 150 nM. To date, no in vivo studies or clinical trials have been published, and its safety profile remains unknown.

Potential applications of hematoxilineozin include use as a chemical probe for kinase research and as a scaffold for developing anticancer agents. Its stable aromatic framework and functionalizable positions allow for further derivatization, which could lead to compounds with improved potency and selectivity. Researchers interested in medicinal chemistry have cited hematoxilineozin as a promising candidate for structural optimization.

The literature on hematoxilineozin is limited to the initial synthesis report and a handful of computational docking studies.
