Fluorosubstituted refers to chemical compounds in which one or more hydrogen atoms in a parent structure have been replaced by fluorine. This term is used across organic, organometallic, and inorganic chemistry to describe molecules containing fluorine substituents on rings, chains, or heteroatoms, such as fluorobenzenes, fluoroalkanes, or fluorinated heterocycles.

Fluorine’s small size and high electronegativity give the C–F bond distinctive properties. The bond is relatively strong and often chemically inert under many conditions, while fluorine can strongly influence the electronic environment of the molecule. Introducing fluorosubstituents can decrease basicity, alter acidity (pKa), increase metabolic stability, and modify lipophilicity and polarity. In aromatic systems, fluorine can withdraw electron density through induction, affecting reactivity and interactions with biological targets or materials. In aliphatic contexts, C–F bonds can influence conformations and steric profiles.

Fluorosubstituted motifs are prevalent in pharmaceuticals, agrochemicals, and materials science because they can tune pharmacokinetic properties, binding affinity, and stability. Examples range from simple fluorobenzene derivatives to multifunctional motifs such as difluoromethyl and trifluoromethyl groups (CF3) attached to various backbones.

Synthesis of fluorosubstituted compounds often employs direct fluorination with electrophilic fluorinating reagents (for example, Selectfluor or NFSI), nucleophilic fluorination of leaving groups, or deoxyfluorination of alcohol derivatives. Regio- and chemoselectivity are important considerations, and specialized reagents or conditions are used to achieve the desired fluorosubstitution. Fluorination is also employed in radiolabeling with isotopes such as fluorine-18 for imaging applications.
