Optimization of elution conditions often involves systematic screening and, increasingly, experimental design approaches to maximize resolution and minimize run time while ensuring robustness. Methods are then validated for precision, accuracy, linearity, limits of detection, and resilience to small deliberate variations in pH, temperature, or flow.
