ESCRTkomponenttien, or ESCRT components, are essential protein complexes involved in cellular processes such as multivesicular body (MVB) formation, protein sorting, and viral budding. The ESCRT machinery is a dynamic system composed of several distinct complexes, primarily ESCRT-0, ESCRT-I, ESCRT-II, and ESCRT-III. These complexes work sequentially and cooperatively to recognize, bind, and engulf ubiquitously tagged proteins destined for degradation or sorting.

ESCRT-0 is the initial recognition module, binding to ubiquitinated cargo. It then recruits ESCRT-I, which further stabilizes the cargo and begins the process of membrane deformation. ESCRT-II, in turn, interacts with ESCRT-I and further bends the membrane, preparing it for the action of ESCRT-III. ESCRT-III is the core machinery responsible for membrane scission, the final step in the formation of intraluminal vesicles (ILVs) within MVBs. Accessory proteins, such as Vps4, are crucial for the disassembly of ESCRT-III complexes, allowing for their recycling and continued function.

Beyond MVB formation, ESCRT components play roles in cytokinesis, the process of cell division, and in the release of enveloped viruses from host cells. Their ability to manipulate and deform cell membranes makes them versatile players in a variety of cellular events. Dysregulation of ESCRT component function has been linked to various diseases, including cancer and neurological disorders, highlighting their fundamental importance in cellular homeostasis.
