Depyrogenation is a process used to remove pyrogens from solutions, typically in pharmaceutical and medical applications. Pyrogens are fever-inducing substances, most commonly bacterial endotoxins, which can cause severe adverse reactions if introduced into the body. The primary goal of depyrogenation is to ensure the safety and efficacy of injectable drugs, medical devices, and other sterile products.

There are several methods for depyrogenation. Dry heat sterilization is a widely used technique, where high temperatures (often exceeding 180 degrees Celsius) for extended periods are employed to denature and decompose endotoxins. Another common method is steam sterilization, utilizing autoclaves to achieve high temperatures and pressure, which also effectively destroys pyrogens. Filtration is also employed, particularly for heat-sensitive solutions. Specialized filters with very small pore sizes (e.g., 0.01 micrometers) can physically remove endotoxins from the liquid. Chemical methods, such as treatment with strong oxidizing agents, can also be used, though these are less common due to potential interactions with the product. The choice of depyrogenation method depends on the product's characteristics, such as its heat stability and chemical composition. Rigorous testing is performed after depyrogenation to confirm the absence of pyrogens before the product can be released for use.
