Ddimeertest

Ddimeertest is a laboratory assay designed to quantify the tendency of molecules to form dimers in solution. It was developed as a standardized approach for comparing dimerization propensity across proteins and synthetic polymers, providing a consistent readout under defined conditions.

Principle and procedure: In a typical implementation, the analyte is labeled with a pair of compatible detection moieties that generate a measurable signal only when two molecules come into proximity as a dimer. The assay is performed over a range of concentrations and buffered conditions, and signals are converted into a Ddimeer index (DI) using a calibration curve derived from known monomeric and dimeric standards. The DI represents an estimate of the fraction of dimers present under the tested conditions.

Applications: Ddimeertest supports studies in basic biochemistry, protein engineering, and early-stage drug screening, where changes in dimerization influence function or aggregation. In polymer chemistry, it can be used to monitor self-association of monomers in solution and to compare the effects of additives or mutations on dimer formation.

Limitations and interpretation: Results depend on the tagging strategy, assay conditions, and the stability of higher-order oligomers. The method may over- or under-estimate dimer content for transient interactions or for molecules that form larger assemblies. Proper controls are essential, and complementary methods are recommended for confirmation.
