Broadspecificity is a term used to describe the property of a molecule, enzyme, receptor, or biological system to interact with a relatively wide range of substrates, ligands, or targets. It stands in contrast to high specificity, where interactions are restricted to a small set of substrates or targets. The phrase appears in discussions across biochemistry, pharmacology, and immunology, often as an informal descriptor rather than a formal category in nomenclature.

In enzymes and proteins, broadspecificity can arise from structural features such as flexible active sites or adaptable binding pockets that accommodate diverse substrates. In receptors or binding proteins, broad binding specificity may result from flexible recognition elements or broad affinity profiles. In pharmacology, broadspecificity can describe drugs or agents with polypharmacology—the ability to affect multiple molecular targets rather than a single modality. In antimicrobial research, broad-spectrum activity is a related concept that captures a drug’s effectiveness against a wide range of organisms.

Measuring broadspecificity typically involves substrate scope profiling, where an entity is tested against a panel of substrates or targets to gauge the breadth of activity. Metrics may include the number of substrates recognized, range of binding affinities, or catalytic efficiency across substrates. This property can offer advantages such as versatility and resilience to mutation or variation, but also poses risks, including off-target effects and increased potential for adverse outcomes.
