Trimethylsilyl is a chemical functional group with the formula (CH3)3Si-. It is often abbreviated as TMS. This group consists of a silicon atom bonded to three methyl groups and one other atom or group. The trimethylsilyl group is commonly encountered in organic chemistry and is widely used as a protecting group for hydroxyl, amino, and other functional groups. Its bulky nature and relative inertness make it effective at shielding these functionalities during chemical reactions.

The trimethylsilyl group can be introduced into molecules through various silylation reactions, typically using reagents like trimethylsilyl chloride (TMSCl) or hexamethyldisilazane (HMDS) in the presence of a base. The stability of the Si-C bond in the TMS group allows it to withstand a range of reaction conditions. However, it can be selectively removed, or deprotected, using agents such as fluoride ions (e.g., tetrabutylammonium fluoride, TBAF) or acidic conditions, regenerating the original functional group.

Trimethylsilyl ethers, formed by silylating alcohols, are particularly common. These protected alcohols are soluble in organic solvents and can be readily transformed into other derivatives. In analytical chemistry, the trimethylsilyl group is often used to increase the volatility and thermal stability of compounds for gas chromatography (GC) and mass spectrometry (MS) analysis, making it easier to detect and identify them. Its widespread utility stems from its ease of introduction, stability, and facile removal.
