Toxoids are inactivated toxins used in vaccines to stimulate protective immunity without causing disease. They are produced by detoxifying bacterial toxins, most commonly the diphtheria toxin and the tetanus toxin, through chemical treatment or controlled heating. The detoxification process preserves the toxin’s antigenic structure, allowing the immune system to recognize it and develop targeted antibodies.

When a person is vaccinated with a toxoid, the body generates neutralizing antibodies that specifically bind to the toxin. If later exposure to the toxin occurs, these antibodies prevent the toxin from damaging tissues, thereby reducing or preventing disease symptoms.

Toxoid vaccines are non-live and typically require booster doses to maintain immunity, since antibody levels can wane over time. They are often formulated with adjuvants and used in combination vaccines. The best-known examples are diphtheria toxoid and tetanus toxoid vaccines, which are widely used in combination vaccines such as DTaP in children and Td/Tdap in adolescents and adults.

Limitations include that toxoid vaccines protect against disease caused by a toxin rather than infection by the bacterium itself, and they may not induce strong mucosal immunity. Production must ensure complete detoxification to avoid disease while preserving immunogenicity, making quality control essential in manufacturing.
