Substituentie is a term used in chemistry to refer to an atom or group of atoms that is attached to a parent molecule by replacing one or more hydrogen atoms. In organic chemistry, substituents define how a core framework is modified and thereby influence the molecule’s properties and reactivity.

Substituents vary widely and include alkyl and aryl groups, halogens (such as fluorine, chlorine, bromine, iodine), and functional groups such as hydroxyl, amino, nitro, carbonyl, and carboxyl groups. They can be broadly classified as electron-donating or electron-withdrawing, acting through inductive or resonance effects to alter acidity, basicity, polarity, and reaction rates.

Nomenclature treats substituents as prefixes to the parent structure, with locants indicating their positions. When multiple substituents are present, numbers are chosen to give the lowest possible sequence. On aromatic rings such as benzene, substituent positions are often described using ortho-, meta-, and para- nomenclature or explicit numerical locants.

Examples illustrate the concept: substituting a hydrogen in methane with chlorine yields chloromethane (CH3Cl); substituting a hydrogen on benzene with a chlorine atom yields chlorobenzene (C6H5Cl); and methoxybenzene (anisole, C6H5OCH3) contains a methoxy substituent. Substituents affect physical properties like boiling and melting points and solubility, as well as chemical reactivity, including directing effects in electrophilic aromatic substitution reactions.

Related to substituentie is the substitution reaction, a class of reactions in which a substituent is introduced by replacing a leaving group. Substituentie thus serves as a descriptive link between molecular structure, properties, and reactivity.
