Steroid receptor types refer to a family of intracellular proteins that bind to steroid hormones and then regulate gene expression. These receptors are crucial for mediating the effects of steroid hormones like cortisol, aldosterone, estrogen, progesterone, and androgens. The steroid receptor superfamily is a large group of nuclear receptors that share structural similarities, including a DNA-binding domain and a ligand-binding domain.

There are several main classes of steroid receptors, often categorized based on their primary function and location within the cell. For instance, Type I receptors, such as the androgen receptor, estrogen receptor, progesterone receptor, and glucocorticoid receptor, are typically found in the cytoplasm in an inactive state, bound to heat shock proteins. Upon binding of their respective steroid ligand, they undergo conformational changes, dissociate from the heat shock proteins, and translocate to the nucleus. Type II receptors, like the thyroid hormone receptor and retinoic acid receptor, are generally found in the nucleus, already bound to DNA, often in complex with corepressor proteins. Ligand binding leads to a conformational change that displaces the corepressors and recruits coactivators, thereby modulating gene expression. While technically nuclear receptors, their mechanism of action differs from Type I receptors. Understanding these different types of steroid receptors and their mechanisms is fundamental to comprehending how steroid hormones influence a vast array of physiological processes, including development, metabolism, reproduction, and immune function. Dysregulation of steroid receptor activity is implicated in numerous diseases, making them important targets for pharmacological interventions.
