Proteintranslokoner, or protein translocons, are protein-conducting channels embedded in cellular membranes that mediate the movement of polypeptides across or into membranes. They are central to protein sorting, secretion, and membrane integration, enabling both secreted proteins and membrane proteins to reach their proper destinations.

Across organisms, several translocon systems exist. In bacteria, the SecYEG complex forms the core channel in the inner membrane, with the SecA ATPase providing the energy to drive post-translational translocation. In eukaryotes, the Sec61 complex constitutes the ER translocon, handling co-translational translocation in collaboration with the ribosome and SRP, and working with accessory factors such as TRAP, OST, and signal peptidases. In mitochondria and chloroplasts, specialized translocons such as TOM/TIM and TOC/TIC facilitate import of nuclear-encoded proteins across the outer and inner membranes of these organelles.

Mechanistically, translocation can be co-translational, where the ribosome connects directly to the translocon as the polypeptide emerges, or post-translational, where soluble chaperones keep the polypeptide unfolded and deliver it to the channel. Energetics vary: bacterial SecA relies on ATP; mitochondrial and chloroplast routes often use a combination of membrane potential and matrix chaperones with ATP hydrolysis.

Significance and evolution: proteintranslokoner are essential for proper protein localization, organelle biogenesis, and cellular function. They are highly conserved yet diversified to meet organelle-specific needs, reflecting the central role of protein sorting in cellular organization. Defects in translocon components can disrupt secretion, respiration, and membrane protein assembly.
