Proteintimport is a cellular process by which proteins are transported from their site of synthesis to specific intracellular compartments or across lipid membranes. It relies on targeting signals, receptor proteins, and membrane translocases to recognize cargo and mediate passage, often coupled to energy sources such as ATP hydrolysis or ion gradients.

In mitochondria, most proteins are synthesized in the cytosol and imported via the translocase of the outer membrane (TOM) followed by the translocase of the inner membrane (TIM). Cargo proteins pass through these channels in an unfolded or chaperone-assisted state, are processed by peptidases, and are folded by mitochondrial chaperones after reaching their destination.

Chloroplasts use the TOC/TIC system to import nuclear-encoded proteins. Precursors bearing chloroplast transit peptides are recognized by outer-m membrane receptors and threaded through translocons into the stroma or thylakoid lumen, often requiring ATP and a proton motive force for translocation and maturation.

Other major routes include import into the endoplasmic reticulum via signal peptides and the Sec61 translocon (with co- or post-translational targeting), peroxisomes via PTS signals and PEX proteins, and the nucleus through nuclear localization signals, importins, and the nuclear pore complex. Each pathway involves specialized receptors, translocation channels, and quality-control steps to ensure correct localization and folding.

Regulation of proteintimport is tightly coordinated with cellular physiology and development, and defects can contribute to disease or impaired organelle function. Proteintimport remains a central focus in cell biology and biotechnology, informing approaches to protein engineering and therapeutic delivery.
