Oxygenresponsive is an adjective used in biology and biotechnology to describe systems, genes, promoters, and devices that detect and respond to oxygen availability. These mechanisms enable organisms and engineered constructs to adapt to hypoxic (low-oxygen) or hyperoxic (high-oxygen) conditions by altering gene expression, metabolism, or behavior.

In bacteria and archaea, oxygen sensing commonly involves transcription factors whose activity depends on the redox state of iron-sulfur clusters. A classic example is FNR in Escherichia coli, which remains active under anaerobic conditions and activates genes for anaerobic respiration; exposure to oxygen causes disassembly of its Fe-S cluster and inactivates it. Other sensors, like ArcB/ArcA, convey redox information to the transcriptional network, adjusting respiration accordingly.

In animals and fungi, oxygen-responsive regulation is prominently mediated by hypoxia-inducible factors. HIF-1α is hydroxylated in the presence of oxygen by prolyl hydroxylases and targeted for degradation. Under hypoxia, HIF-1α stabilizes, dimerizes with HIF-1β, and drives transcription of genes involved in angiogenesis, glycolysis, and iron metabolism through hypoxia response elements.

In plants, oxygen sensing involves group VII Ethylene Response Factors and the N-end rule pathway. Under normoxia, plant PCO enzymes hydroxylate an N-terminal cysteine, marking the ERF-VII transcription factors for degradation; hypoxia stabilizes these factors to reprogram metabolism and development.

Applications include oxygen-responsive promoters and reporters used in research and industrial biotechnology to control expression based on oxygen tension. They are employed to optimize fermentation, study hypoxia biology, or construct synthetic gene switches. Measurement of oxygen responsiveness relies on sensors and reporters, such as pO2 probes or reporters driven by HRE or ARE elements.
