Lichtmicroscopen, also known as optical microscopen, are instruments used to magnify small objects and structures that are invisible to the naked eye. They work by passing light through a specimen and then through a series of lenses. The primary lenses, the objective lens and the eyepiece lens, work together to create a magnified image. The objective lens, located near the specimen, provides the initial magnification, while the eyepiece lens further magnifies this image for the observer.

The resolution of a light microscope, its ability to distinguish between two closely spaced points, is limited by the wavelength of light used. Typically, light microscopes can achieve magnifications up to around 1000 times, with a resolution limit of about 200 nanometers. Different types of light microscopy exist, each employing variations in illumination and contrast techniques to visualize specific features. Common types include bright-field microscopy, dark-field microscopy, phase-contrast microscopy, and fluorescence microscopy.

Bright-field microscopy is the most basic form, where light passes directly through a stained or naturally contrasting specimen. Dark-field microscopy illuminates the specimen from the sides, making it appear bright against a dark background, useful for unstained transparent samples. Phase-contrast microscopy enhances contrast in unstained, transparent specimens by converting phase shifts in light into amplitude changes. Fluorescence microscopy utilizes fluorophores, which emit light of a specific color when excited by light of another color, allowing for the visualization of specific molecules or structures within cells.
