LeucinZipper is a synthetic protein motif proposed to form a leucine-rich coiled-coil dimerization domain, analogous to the natural leucine zipper but distinct in sequence and origin. It is used in modular protein engineering to drive specific dimerization of effector proteins.

Structure: LeucinZipper modules rely on alpha-helical coiled-coils with heptad repeats (abcdefg)n in which leucine or similar hydrophobic residues occupy the d position. The regular, hydrophobic "zipper" interface stabilizes dimers and can be tuned to favor homodimeric or heterodimeric pairings through sequence variation.
