Glutathione peroxidase is a family of enzymes that play a crucial role in protecting cells from oxidative damage. These enzymes catalyze the reduction of hydrogen peroxide and organic hydroperoxides, using reduced glutathione as a substrate. This process converts harmful reactive oxygen species into water, thereby preventing cellular damage.

There are several isoforms of glutathione peroxidase, with different subcellular localizations and substrate specificities. Glutathione peroxidase 1 (GPx1) is the most abundant and is found in the cytoplasm of most tissues. It primarily reduces hydrogen peroxide. Glutathione peroxidase 4 (GPx4) is unique in that it can reduce lipid hydroperoxides, protecting cell membranes from oxidative damage. Other isoforms, such as GPx2, GPx3, and GPx6, have more specialized roles and tissue distributions.

The activity of glutathione peroxidases is essential for maintaining cellular redox balance and is linked to various physiological processes, including defense against inflammation, detoxification, and immune responses. Deficiencies or altered activity of these enzymes have been implicated in the pathogenesis of several diseases, including cancer, cardiovascular disease, and neurodegenerative disorders. Selenium is a critical component of the active site of glutathione peroxidases, and its deficiency can impair enzyme function.
