Glutamincykeln, often referred to as the glutamate-glutamine cycle, is a metabolic pathway crucial for neuronal function in the central nervous system. It involves the neurotransmitter glutamate and its precursor glutamine. Glutamate is a primary excitatory neurotransmitter, playing a key role in learning, memory, and synaptic plasticity. However, excessive extracellular glutamate can be excitotoxic, leading to neuronal damage.

The cycle begins with the release of glutamate from presynaptic neurons into the synaptic cleft. It then binds to postsynaptic receptors, triggering a signal. To terminate this signal and prevent excitotoxicity, glutamate is rapidly cleared from the synaptic cleft. This clearance is primarily mediated by excitatory amino acid transporters (EAATs), which are located on both neurons and glial cells, particularly astrocytes.

Astrocytes play a central role in the glutamate-glutamine cycle. They take up a significant portion of the released glutamate via EAATs. Inside astrocytes, the enzyme glutamine synthetase converts glutamate into glutamine. This conversion is vital for both recycling the neurotransmitter and detoxifying excess glutamate. Glutamine is a relatively inert molecule and can be safely transported.

Astrocytes then release glutamine back into the extracellular space. Neurons possess another transporter, the system N transporter, which imports glutamine from the extracellular space. Once inside the neuron, the enzyme glutaminase converts glutamine back into glutamate, thus replenishing the neurotransmitter pool for subsequent synaptic release. This continuous cycling ensures the availability of glutamate for neurotransmission while protecting neurons from glutamate-induced damage. Disruptions in the glutamate-glutamine cycle have been implicated in various neurological disorders, including epilepsy, stroke, and neurodegenerative diseases.
