Endothelsynthase is a term that appears in some speculative discussions of endothelial metabolism to denote a proposed enzyme that would catalyze the biosynthesis of certain signaling molecules within the vascular endothelium. It is not part of established enzyme nomenclature, and there is no widely cited primary literature confirming its existence as a defined enzyme with a specific reaction in humans or other organisms. Consequently, it does not have a canonical entry in major biochemical databases.

In proposed models, endothelsynthase would be a synthase or transferase localized to intracellular membranes of endothelial cells, potentially the endoplasmic reticulum or Golgi apparatus. Its suggested function would be to assemble precursor molecules into bioactive endothelial mediators that participate in autocrine or paracrine signaling to regulate vascular tone, inflammation, or angiogenesis. Because the exact substrates, products, and catalytic mechanism are not established, any description remains hypothetical.

Relation to real endothelial enzymes: the term should not be confused with endothelin-converting enzyme (ECE) or endothelial nitric oxide synthase (eNOS), both of which are well characterized.
