Denosumab is a medication used to treat several bone-related conditions. It is a human monoclonal antibody that targets RANKL, a protein that plays a crucial role in the formation, function, and survival of osteoclasts. Osteoclasts are cells responsible for breaking down bone. By inhibiting RANKL, denosumab reduces osteoclast activity, thereby decreasing bone resorption.

Denosumab is primarily prescribed for the treatment of osteoporosis in postmenopausal women at high risk of fracture. It is also used to treat bone loss in men taking certain hormone-blocking therapies for prostate cancer, and in women undergoing adjuvant treatment for breast cancer. Additionally, it is indicated for the treatment of giant cell tumor of bone in adults and skeletally mature adolescents, and for the prevention of skeletal-related events (such as fractures, spinal cord compression, and the need for radiation or surgery to bone) in patients with bone metastases from solid tumors.

Denosumab is administered as a subcutaneous injection. Common side effects can include pain in the extremities, muscle pain, and eczema. A rare but serious side effect is osteonecrosis of the jaw, which involves damage to the jawbone. Hypocalcemia, or low calcium levels in the blood, can also occur and requires monitoring and supplementation.
