Cysteinerich, or cysteine-rich, is a descriptor used in biochemistry and molecular biology for proteins, peptides, and regions of proteins that contain a high proportion of the amino acid cysteine. Cysteine residues feature thiol groups capable of forming disulfide bonds, which can link different parts of a molecule or different subunits and greatly influence structure and stability.

In cysteine-rich molecules, many cysteines participate in disulfide bonds, producing compact, stable folds. These bonds often enhance resistance to proteolysis and harsh environments, a common feature of extracellular and secreted proteins. Cysteine-rich sequences are frequently found in defensins, venom peptides, extracellular matrix components, and certain secretory or membrane-associated proteins.

A distinctive feature of many cysteine-rich proteins is the presence of specific motifs and topologies, such as the cystine knot motif. The cystine knot, formed by a set of disulfide bonds that tie the peptide into a robust topology, is observed in a broad range of toxins, growth factors, and other secreted peptides. Knottins, a related category, describe peptides with particularly compact, disulfide-rich scaffolds.

Examples of cysteine-rich proteins include plant and animal defensins, cysteine-rich secretory proteins (CRISPs) in vertebrates, and various venom peptides such as conotoxins and certain scorpion or spider toxins. These proteins often serve roles in defense, signaling, or interaction with other organisms, and their stable folds make them attractive as scaffolds for peptide engineering.

In research and applications, cysteine-rich motifs are studied for structure-function relationships, proteomic annotation, and as foundations for designing stable peptide-based therapeutics and biotechnological tools.
